Urea in the sample is hydrolized enzymatically into ammonia (NH4
+) and
carbon dioxide (CO2).
Ammonia ions formed reacts with -ketoglutarate in a reaction catalysed by
glutamate dehydrogenase (GLDH) with simultaneous oxidation of NADH to
NAD+
